Our ability to "read" DNA has made tremendous progress in the past few decades, but the ability to understand and alter the genetic code, that is, to "rewrite" the DNA-encoded instructions, has lagged behind. A new Weizmann Institute study advances our understanding of the genetic code: It proposes a way of effectively introducing numerous carefully planned DNA segments into genomes of living cells and of testing the effects of these changes. The study is being reported in the June issues of Nature Biotechnology and Nature Genetics.

Until now, changing the DNA sequence has been a slow and labor-intensive process. It took several weeks to alter just one DNA region at a time; testing the effects of each of these changes took even longer. In the new study, Weizmann Institute scientists have developed a technology that makes it possible to simultaneously introduce tens of thousands of DNA regions into tens of thousands of living cells ? each region in a separate cell ? in a planned and systematic manner, and to measure the results of each such change with great precision and within a single experiment.

"This fast method will significantly advance our ability to understand the 'language' of DNA," says research team leader Prof. Eran Segal, of the Weizmann Institute's Computer Science and Applied Mathematics and Molecular Cell Biology Departments. "Reading out a person's entire genome is already a manageable task, but what exactly is written in that genome? After all, a genome looks like a lengthy string of letters whose meaning is for the most part obscure. Just deciphering the DNA letters is like trying to understand a foreign language by listening to it being spoken. Our method will help us identify DNA 'words' and understand their meaning."

Understanding what's written in the DNA might help us interpret, among other things, how genotypic differences among people generate observable differences among them, from the way we look to the way our cells function. Thus, for example, it might be possible to clarify which genetic differences are responsible for the development of various diseases in certain individuals. The Weizmann Institute technology can also lead to improved genetic therapies based on introducing new genes or improved regulatory sequences into cells in order to repair genetic defects.

In the present study, the scientists investigated a vital aspect of the DNA language: How the control of gene expression is encoded in the DNA ? that is, the instructions determining the level of activity of each gene in the genetic code. Since gene activity levels have crucial effects on cell function, this question, considered one of the central in molecular biology, has been studied for decades. The new technology has enabled the scientists to isolate and test the effects of various parameters on gene activity levels: For example, how a gene's activity level is affected by the gene's distance from its regulatory sequence. The researchers have managed to elucidate how various parameters define the regulatory "language" and to demonstrate how deliberate changes in the genetic sequence affect these parameters in a way that alters the level of a gene's activity in a predictable manner.

The new method consists of four steps that combine existing technologies in an innovative manner. The steps are: creation of 50,000 different genetic sequences on DNA chips; massive insertion of these sequences into cells at the same time; sorting the cells with the help of a sorting machine that senses the expression levels of a "reporter" gene; and high-throughput parallel DNA sequencing.

Taking part in the study were Weizmann Institute's graduate students Eilon Sharon, Tali Raveh-Sadka and Michal Levo, research assistant Dr. Yael Kalma and research associate Dr. Adina Weinberger, as well as Dr. Zohar Yakhini from the Technion ? Understands that no skill or talent can overshadow the act of preparation.
Capable of managing temper and anger.
Keeps other people?s feeling in mind and limits selfishness.
Being able to distinguish between ?needs? and ?wants?
Shows confidence without being overly arrogant.
Handles pressure with self composure.
Takes ownership and responsibility of personal actions.
Manages personal fears.
Able to see the various shades of grey between the extremes of black and white in every situation.
Accepts negative feedback as a tool for self improvement.
Aware of personal insecurities and self-esteem.
Able to separate true love from transitory infatuation.
Understanding that open communication is the key to progression.
Above all, true adults do what they have to do when it is required of them, and they do what they want when they can. They are able to distinguish between the two and manage their time and efforts accordingly.

Anti-Psychotic Drugs Found to Bust Cancer Cells

An anti-psychotic drug could treat cancer by neutralising the tumour's growth. Scientists from Cancer Research UK and McMaster University in Ontario have found that an anti-psychotic drug called thioridazine has the capability to cure cancer. The drug transforms cancer stem cells into non-cancerous cells, they have found.

Many researchers believe that cancer stem cells are the source of new cancer cells within a tumour. They claim that these cancer stem cells were resistant to current treatments, which explains why some cancers are so difficult to cure.

To find a cure for cancer stem cells, scientists had used pioneering robotics to test more than a dozen different compounds to identify a drug which could cure or eradicate the cancer stem cells. They found that the anti-psychotic drug does not eradicate the cancer stem cells but converts the cancer stem cells into non cancerous cells, that too, without damaging healthy cells.

"The unusual aspect of our finding is the way this human-ready drug actually kills cancer stem cells by changing them into cells that are non-cancerous," said Dr Mick Bhatia, scientific director at the McMaster's Stem Cell and Cancer Research Institute.

"Now we can test thousands of compounds, eventually defining a candidate drug that has little effect on normal stem cells but kills the cells that start the tumour," he added.

Researchers are planning a clinical trial of thioridazine in combination with standard anti-cancer drugs for adults with acute myeloid leukaemia.

"It's too early to say whether thioridazine could be used to treat cancer patients, but the research opens up some interesting questions for further investigation. Use of thioridazine at dose levels required to treat cancer may well result in significant side effects that limit or prohibit its use. Further research will be needed before we can be sure," said Dr Tim Somervaille, researcher at Cancer Research UK, in a statement.

Celldex scores upbeat data for breast cancer drug - FierceBiotech

Celldex Therapeutics ($CLDX) has provided positive data for its targeted drug against tough-to-treat breast tumors, and executives expect to seek a nod from regulators to advance the experimental treatment into a late-stage study required for approval.

The Needham, MA-based company's CDX-011 consists of an antibody targeted to the GPNMB protein and an attached chemotherapy agent that is supposed to hit tumor cells with a killer punch. In a 122-patient study, 19% of patients on Celldex's drug had a response compared with 14% among patients who got standard chemotherapy. And investigators found that response rates were higher in patients with increased levels of GPNMB, a protein on tumor cells that enables cancer to mobilize and spread. Prior to the study, all participants had received previous treatments.

The data from the mid-stage trial are preliminary and don't shed much light on the survival benefit of the treatment, yet Celldex is planning to release further details late this year. Executives aim to get the FDA to sign off on an advanced study of the drug to combat breast tumors that express GPNMB and fall into the triple-negative category, TheStreet reported. These difficult-to-treat tumors account for about a third of cases of breast cancer, according to the company, which acquired the drug in its 2009 buyout of CuraGen. Celldex has a license to antibody linking technology and cell-killing agent for CDX-011 from Seattle Genetics ($SGEN), which uses the same tech for its approved lymphoma drug Adcetris.

"GPNMB helps the tumor cell to move around," said Dr. Linda Vahdat, director of the breast cancer research program at Weill Cornell Medical College and the lead investigator of the study, as quoted by Reuters. "It almost seems like this drug puts the brakes on that process by taking away its legs."

As Adam Feuerstein at TheStreet noted, however, Celldex hasn't provided any proof that CDX-011 prolongs survival or stymies re-growth of tumors for a significant period of time. And those are the types of endpoints that regulators in the U.S. and Europe like to see programs reach before green-lighting cancer drugs.?
